General description
Transcription factor GATA-6 (UniProt: Q92908; also known as GATA-binding factor 6) is encoded by the GATA6 gene (Gene ID: 2627) in human. GATA6 is a zinc-finger transcription factor that belongs to the six-member family of transcription factors that bind to the consensus sequence (A/T)GATA(A/G). GATA1, GATA2, and GATA3 are mainly expressed in hematopoietic cell lineages, while GATA4, GATA5, and GATA6 are predominantly expressed in the heart, gonads, and endodermal-derived tissues. It has two zinc-finger repeats localized to amino acids 390-414 and 444-468, which mediate interaction with LIM and cysteine-rich domains protein 1 (LMCD1). GATA-6 is shown to be important in the endodermal differentiation of organ tissues and has been indicated in proper lung development by controlling the late differentiation stages of alveolar epithelium and aquaporin-5 promoter activation. It has also been linked to the production of LIF that encourages proliferation of endodermal embryonic stem cells and blocks early epiblast differentiation. GATA6 is also required for definitive endoderm specification in human embryonic stem cells and induced pluripotent stem cells. Phosphorylation of GATA6 by GSK-3 on serine 37 is reported to promote its degradation, thereby preventing GATA6 from repressing transcripts that are induced by TNF and attenuated by insulin.
